When a certain רב was yet a child, he said that it is well known that a relative is פסול לעדות even לחובה.   “But why?” he asked.  “He’s not נוגע בדבר, when he’s testifying to the detriment of his relative.”  The child answered his question:  “The תורה says with respect to עדות that ועמדו שני האנשים.  This teaches that they have to be אנשים.   And a person who is willing to testify against his own relative is not בחזקת איש.”
